Can I purchase one day passes to Epcot at the Disney Store?

No, they only sell park hopper passes:

4 day
5 day
5 day plus
6 day plus
7 day plus
annual passes voucher

You can only buy one day passes at the parks themselves.

One day passes can be purchased at all Disney resorts at both the front desk and at Guest Services, at any WDW major park ticket booth, or at any Guest Relations window/office at the parks or at DTD.
