If I purchase air tickets from one of the travel web sites and cannot select my seats at the time of purchase, will I be able to call the airline directly and purchase them?

Maybe yes and maybe no, but it isn't a good idea to purchase airline tickets from a travel website. If anything changes or the flight gets cancelled you won't be able to deal with the airline directly you will need to go through the travel site and they aren't the easiest to work with or get a live person to talk to for that matter.
