We are planning for April. We will be staying offsite for the beginning of the trip. So, I had planned on hiring a car to take us there, but I am planning on staying onsite for the last few days.

Can I use Magic Express one way? Is that allowed? Thanks!

Yep, no problem. You can book it when you make your Disney room reservation...just give them your info for departure day. Or, you can wait until you get to the resort to checkin, and then make your DME deparure reservation when you are at the front desk. You should do it as soon as you check in so that you can be sure you are in the system.
Either way will work though.

Also, if you are renting and returning your car at MCO, you can use ME both ways! Set a reservation for the day your are checking in at Disney. I would assume you would be dropping the car off that day at MCO. Drop the car at MCO and walk to ME. You would then have ME set up for your return trip to MCO. To do the booking from MCO to WDW, simply give them any airline flight that arrives about the time you want to return the car at MCO... you are entitled to one trip from MCO to WDW and one trip from WDW to MCO during your stay.
