We are flying into MCO and taking the DME directly to our WDW resort, and spending 8 nights there. Then we are renting a car, driving to Tampa, and spending 2 nights in Tampa. We are returning to Orlando, and spending one night offsite, just outside WDW, the night before we fly out of Orlando.
Am I able to go back to my resort, and get the DME back to the airport based on my previous stay at a WDW resort 3 nights earlier?

I don't think so, but I could be wrong. I know you can go back early, but I don't think you can go back late. ME is not there for your convenience so much as to convince you that you should stay in a Disney hotel rather than someone else's.

Why don't you just stay at a cheap airport hotel and just return the car to the airport?

No you can't use DME you can use DME once from and once to the airport at anytime while you are staying on property once your reservation is done you can no longer use it.

Pick up and drop off at WDW ...$120
Pick up at WDW, drop off at airport ...$200 (for same time period)

It's less expensive to take a taxi back to airport.

So when checking in for DME at the resort, do they ask to see if you stayed that night at the resort?

You can only use DME during the time you are a WDW Resort guest.

Even though DME is free you must have a confirmed reservation. Since you won't be a guest on your departure day you won't be able to make a reservation. On the way from the resort to the airport guests are scheduled on a particular bus based on resort and flight time. The driver scans your voucher prior to boarding guests on the bus, you won't have a voucher so you won't be able to to board the bus.

You don't just check in for DME at the resort. DME gives you an assigned pick up time based on your flight time. You show up 10 minutes before your assigned bus pickup time. Giving Disney a flight that leaves 3 days after you check out won't work.

Stay at a Disney value resort instead of offsite. A taxi (or car service) will probably cost $65-$75 with tip. I'd just pay the extra couple of dollars.

edited to add--The driver isn't going to check and see if you stayed at the hotel the previous night. The driver is going to check your voucher. You "lose" your voucher? Your name won't be on the list. You'll ask to get your voucher reprinted. You'll be told NO, you check out 3 days ago.
