If you have a TouringPlans subscription, they will send the room request about 30 days out.
From what I have read, whatever the most recent request sent will be what they try to accommodate. Which is why, if you have made a request with TouringPlans or via chat or phone, you shouldn't add anything when you check in online. The room request info, I believe, is made in the notes to the reservation.

From what I understand, the risk with doing online check-in and a separate room request another way, is that if the two don't match exactly, the room assigner (person) doesn't know what to do.
I don't know how the new automated room-assignment system deals with multiple, different request though.
