I am another vote for 4 - 5 days, ideally. Heck, I am hoping to eventually swing a 4 - 5-day trip for myself! Yes, of course, you could do DLR in 3 days. You can even do it 2 days, and you could even have a really full and rewarding 1-day trip to DLR, but if you really like to look at everything - really soak in all the hidden gems and details that make DLR so special, like the weather vanes and the different bathroom signs and the edible plants in Tomorrowland - you need a lesiurely pace. You need to be able to spread out your activites in both parks and get some good rest breaks over several days so nobody gets burnt out. You should take time to roam around all 3 of the DLR hotels and search for Hidden Mickeys and maybe take in a character meal or two or three, and stroll through Downtown Disney, maybe eat at Rainforest Cafe. You need time to look in all the special shops in DLR, and pick up pressed pennies along the way. You need time to explore ToonTown and A Bug's Land fully, and time to experience the Redwood Creek Challenge Trail and the Animation Academy. You need to see the Aladdin musical and Turtle Talk with Crush. You should see Billy Hill over in DL. You need to be able to ride Toy Story Midway Mania 10 times and Buzz Lightyear 8 times! You need to meet lots of characters and sample some of the different restaurants around DLR. You need to have a silhouette and/or a caricature made of your children. And the rides - there are so many other rides to go on in DLR.
Those 4 - 5 days will fly by surprisingly fast. If you are not much into details and really finding the hidden treasures that we all love about DLR, then yes, you could certainly do a 2-day trip or a 3-day trip. But if you can afford to dtay an extra couple of days, I think it would be worthwhile!
