I know that test rack has a single rider entrance but does mission space have it?
 
There is a single rider queue, but IMO it will not be as effective as the single rider queue for Test Track (and MIB at USO). On Sunday, I joined the standby queue which was full up to just inside the doorway. A few people entered the single rider queue at about the same time. I ended up riding well before they did. Because M:S seats 4 guests per pod, there tend to be fewer single seats available when compared to attractions that seat 3 per row.
 
My experience was similar to PammyK's; when we went last Thursday the single rider wait clock said 35 minutes and the standby line said 20 minutes--I asked the CM if this was accurate and she said that, yes, the single rider wait had been running a bit longer than standby. DH went in the single rider line and waited 40 minutes. I went in the standby line right after he rode and waited 15 minutes. I'm not sure if this is something they will work with and tweak before the official opening, as it would seem a shame to "punish" single riders with a longer wait time!
