I flew with mine last weekend. Did ask TSA on outbound and was told it had to be removed from case. I just put it in the bin for return flight screening also.
This is just personal experience. As advised above, the TSA agent may ask you to remove it. I have gone through TSA with an ipad 12 times and have not removed or asked for it to be removed any of those times. 4 times and the same results for the Kindle Fire. I have not heard of many people being asked to remove tablets.
No one's ever asked me to remove my e-reader or my Ipod from my backpack. Once at DCA they wanted to check through the pack because they thought I had too many electrical cords (all those chargers, you know)
