IS there cheaper transportation to Port than DME?

I presume you mean Orlando, and a lot of it depends on how many people are in your party. For our party of four, we have found that a rental car is often cheaper than taking the Disney bus, plus it affords more flexibility.

There are 5 of us. Do you just rent a car at the airport and then is there a place at Port Canaveral to turn it in?

And yes we are flying into MCO

This. For just 1 or 2 people, DCL's price isn't bad. It's if you have more than that that there are less expensive options as the other posters mention. :-)

For 5 people it would be more cost effective to rent a car and turn it in at Port Canaveral then shuttle over to the DCL terminal. Or have the driver drop off all and the luggage, return the rental car and shuttle back to port to board with the folks left at the terminal.
You could check various private car services as well like Carey International of Park Avenue Limo to compare rates. That way they drop you all off and you set the time for drop off and return pick up.

More than 3 you'll most often come out ahead doing 1-way rentals. There is more logistics in doing (CCC on-site can be costly for 1-ways and have always found a lot better rates from MCO, so need to take ME to airport, rent, as PP said drop-off everyone and the luggage at port, drop-off rental and take shuttle back to port) with one having to decide if the savings worth the time/frustration and inconvenience.

Can't emphasize enough if doing 1-way rentals, as PP said and I cannot agree with more in dropping all at port first. Nothing as frustrating as having a group of 5 with luggage waiting for a too busy shuttle at the car rentals off-site trying to get to port. If you are by yourself without luggage, let the rental company employee that is managing the shuttle know you are a single rider without luggage - this has gotten me on the next shuttle to fill-up with an easy rider multiple times.

As well, I will constantly re-reserve vehicles as I see the costs for drop (canceling the existing res after the new res is made). You may as well (though rental rates have gone up considerably the last few years) have a cheaper cost holding onto the rental even with port parking which is the ultimate in convenience as you have total control of the time you arrive and leave.
 
Our last cruise was 2014 with a family of 5. We flew into MCO and picked up a rental car at airport for a one-day visit to WDW then dropped off at Port before cruise. It was definitely cheaper for us. We ended up paying $31.50 for a one way rental to PC and $57.60 for a one way rental to MCO after the cruise versus $70 x 5. It was a mini van which easily carried all of us plus our luggage. I did drop family off at Port with luggage first then took the shuttle to Port solo from car drop off. Was very easy process.

I think I booked and re-booked my res 5 or 6 times as I kept getting cheaper rates. One thing to remember is the tolls. I usually take the DME to WDW and was glad that I had my magic roll of quarters in one of the M&M tubes (hint from another Dis forum) and had no problems. Would also recommend on-line check-in for the rental to avoid the long lines at MCO to fill out the paperwork.

In November, my DH and I will be taking another cruise. Since there are just two of us, we will probably just use DME.

These were the companies at the port last time we cruised. If they have added or taken away any I am sure someone will post.

Payless, Hertz, Budget, Thrifty/Dollar, Avis, National/Alamo/Enterprise

All are not far from the port with shuttles to the port.

Don't forget that you trade convenience and time at the expense of cost. Rental cars and private car hires are about your only other choices. Sometimes it may be better to do 2 one way rentals, or you could do a round trip and pay for parking at the port. Or stay at a hotel in the area. There are a ton of options.

Just remember opportunity cost. Spending 5 hours researching all the options to save $5 might not be the best use of your time either.
 
If DME means Disney Magical Express, it is not even an option to get to Port Canaveral (which is the port I am assuming you are meaning) from Orlando International Airport (or back), as Magical Express is a complimentary transportation between Orlando International Airport and the Walt Disney World resorts for resort guests only.
 
Payless is no longer at Canaveral.

I’m all about a private transport going to port. Door to door service. No stress. I mostly use Happy Limo. If you look up their rates be sure to add a tip to those numbers for the driver. When I travel with my friends (anywhere from 6 to 20-something of us) we typically spend the same as Disney’s transfers or a few dollars more per person but we ride in vehicles like stretch Hummers or party buses. To me it’s much more exciting and we travel on our own time.
