We want to take the fastest, most convenient mode of transportation from Disney World to Universal - a one-day visit to The Wizarding World of Harry Potter, of course.

What is our best option . . . a taxi? If so, how much might this cost round trip? Would a limo/towncar service be better?

I don't mind renting a car to save, but I do not like the hassle of pickup/drop off for only a one-day rental. Our trip is short (only 4 nights), and I don't want to waste any precious WDW park time picking up and dropping off a rental car.
 
Either a cab or a towncar service is your best option. It should run you about $60-70, plus tip though.
Mears runs a shuttle for about $20 pp, r/t, but it isn't going to run on your time...it is much more ballpark type timing.
 
Taxi is quickest most convenient. Town car is nicest. Taxi will be there when you are leaving and just grab the next one in line. Town car you have to set up a pick up time. Not a huge deal, but you just need a bit of planning involved.

I used Magic Kingdom as a start point and it came to $41.25 before tip.

Happy Limo will be $52.00 ($93.00 round trip) before gratuity. So it will be about $10 more for a private town car. You just need to prearrange pick up times with the car service vs. grabbing a cab at the resort and park. Another benefit I just thought of is I don't know what it is like at closing time to grab a cab from US. A car service will be there just for you - just a thought. Good luck.
