Yes they take Mastercard. In the US, the two are pretty much interchangeable - it is so rare to find a merchant that takes one and not the other that I cannot even recall the experience.
I'm guessing OP is asking because Disney's branded CC is a Visa and they might be thinking of it in terms of "Disney contracts with Coke, so you can't buy Pepsi."

Interestingly, at the London 2012 Olympic Games, only one of the two was accepted as payment for tickets. I can't remember which, whichever one sponsors the Olympics, but it can happen that only one card is viable... Fortunately, that's not a problem at WDW!

You must not shop at Sam's Club (they take MasterCard, but not Visa). Other than that, I can't think of a single merchant who takes one but not the other.
