Closest Costco?

Depends on when, how you want to get there. The closest is probably the one on 441 (Orange Blossom Trail). It is easy to hit if you are coming from the Airport by car and is the closest to ASSp as the crow flies. If driving into Orlando there is one right off the I-4 near Millenia. Access wise, it is probably the best.

If you don't have a car, neither are very close or convenient to Disney.

Just go to Google maps and type in "Costco Orlando" to see where the two are located. The Store Locator on Costco.com will also pull up the options, along with a description of available services at the locations.

The closest one would be the one at the Millenia Mall off of Conroy. There is not one on the south end or Kissimmee.

And the really odd thing is that Google Earth shows the one on Millenia being in the middle of a lake (same if you hit it from the satellite view on the Costco site) even though the Costco site says it was opened in something like '86. If you go to street view, it clearly shows it under construction though. Considering how out of date street view usually is, I can only assume that it is complete and all the imaging is outdated. The one off OBT is quite obviously a Costco with a gasbar in the satellite image on G-Earth, although the Costco site says it only opened last year.

The Costco on OBT closed awhile ago. The Millennia Costco is closest to the parks.

Dug into this a little more (google reviews), and maybe this explains the 2016 opening date. It appears that the standard Costco on OBT converted to a Costco business centre last year (not actually closed). It is still open to all members, but some of the things you would expect at a normal Costco won't be there. For instance, they will probably have restroom paper towels, not kitchen paper towels. No hot foods are available. Some sizes are even bigger than the already huge sizes that regular Costco carries. So probably not the best choice for a vacation stock up. However, the gas bar is still open, and apparently rarely busy if you want cheap gas, and the store itself is much quieter now, so parking is a breeze. Just relaying in summary what a bunch of recent reviewers stated.

Presumably the obviously new one on Millenia has a 1986 opening date listed because they "moved" the OBT location there.

Sorry if my initial response caused confusion. I knew about the new one on Millenia, didn't realise it was a move from OBT.

I visit Orlando twice a year for ten day visits and visit Costco primarily for the cheap gasoline. I have never encountered the Costco gas station on OBT to be closed. It is my last stop before I turn in my rental car at MCO. I enjoy the new Costco Millennia location behind Ross Dress for Less, because it is closer to my timeshare.
