Anyone driven this ?

Best route, time etc

looks to be around 1hr 30mins so presumably do able straight of the plane ?

Yes it's really easy - as you're heading west you avoid most of the Miami traffic. The best way is the US 41 Tamiami Trail, and it's a nice scenic drive across the Everglades. We often go to Sanibel Island, and it's the way we go.
I'd say it would take closer to 2 hours though - it's 100ish miles...

We've been on Tamiami previously, but were thinking of taking Alligator Alley this time to Fort Myers - does anyone have any feedback on this route?

Alligator Alley is quicker than the Tamiami however....watch out for traffic cops (State Troopers?) as I have seen loads of cars getting pulled for speeding.

It is also less scenic

We drove Alligator Alley last year - it's a very boring road as it's very long and straight! But an easy drive :thumbsup2 Keep your eyes peeled for alligators in the water at the side of the road!

For speed and safety, Alligator Alley (I-75) is the way to go. The toll is $2.50 each way, $2.00 if you have a SunPass transponder. The road is boring but it's the fastest way across.
