My sister-in-law is joining my daughter and I for a few days at the Poly. She is on our resort reservation but has her own MDE. (We are “friends” on MDE.). She linked her park ticket to her MDE account, but does she also need to link my resort reservation to her account so I can make fast passes for her?

No; she’s already linked to you and with the reservation including her in the listing, you'll be able to book FPs for everyone.
