We originally setup ME for getting us to and from Old Key West resort. Since setting it up, our waitlist came through and we'll be splitting our stay starting at OKW and ending at the Polynesian. Do I need to call ME and notify them of the change?

And I suspect you now have two different reservation numbers, one for the OKW stay and one for the Poly stay. You would want to add the resort to airport trip under the Poly reservation, since DME is only available from day of arrival to day of departure for a reservation. Your OKW reservation will have already ended by the time you want to return to the airport.
