As many trips as we have taken, I don't remember ever seeing a person in a wheelchair being accommodated on the bus. Question; do people have to be able to walk onto the bus in order to use the magic express?

Of course they can take wheel chairs! You should let them know before (and confirm the day before, I believe in overkill) hand so that you aren't waiting a long time for a bus on arrival. I would also confirm at the front desk or by calling magic express the day before you leave to make sure the wheelchair lift is noted on the reservation. Not every bus is equipped with lift so it's important especially on the way home to make sure they know
