In your opinion, what is the least fulfilled room request?

And yet somehow people manage to pick their own room on cruises. Including the Disney ones!

That's because cruise ships run on a radically different model than hotels.

First, and most importantly, every single passenger on the cruise is going to start on the same day and end on the same day for the entire length of the cruise. While specific boarding/disembarking times may differ due to capacity constraints of getting that many people on one ship at or near the same time, the cruise line knows that for that entire cruise journey every guest is locked in for the entire go. Even if the ship picks up a few passengers at certain ports of call, the turnover is exponentially less. Compare that to hotels where people are staying for various lengths of time and checking in/out at different times (which aside from obligated check in/check out times are not within the control of the hotels) that becomes a far more difficult problem for a hotel. It probably wouldn't be a huge issue if only a few people were able to do it, but if every guest is able to request specific rooms and Disney guarantees that or some kind of compensation it would be an absolute nightmare for them. It would almost certainly result in underused rooms. Since everyone going has different schedules and dates, there would almost certainly be a situation where it would be very hard to lock down a specific room for longer than 2 or 3 days without Disney having to hold rooms back for visitors with longer stays.

The other big difference is maintenance. Unlike cruise ships, which usually have time off between trips for thorough maintenance and cleaning, hotels operate constantly 24/7. That means that rooms often have to be taken offline unpredictably due to maintenance issues. For example, if you booked a certain room and the day before you were supposed to check-in there was a plumbing issue that flooded the room, they would likely have to take that room off-line and put you in a new room. Once again, that might work if only a few people could reserve specific rooms, but it would create a massive trickle-down disaster if everyone was allowed to do it.
 
We are Preferred at POP - that would be '50's, '60's or '70s - any are fine with us, even though DH likes the 1950's
My 1st request is an end room
Then my 2nd request will be upper floor (we would like floor #4)

But an end-room is our #1 request

Most likely request to be granted: whatever is available
 

I would think the least fulfilled request would be the most specific one.

I try to make a very general request (i.e. upper floor) so I have a better chance of getting what I want.
 
Now that me and DH are empty nesters, the only request I put in is not close to elevator. We prefer a longer walk and have it more quiet, no one running by the room.

You definitely can. It's not quite as close as it looks on the small map, it's almost equidistant to the two food courts depending on which end of Mighty Ducks you're on, but still.
That said, the only request we've ever gotten fulfilled was our one-night stay at ASMo when we asked for a Dalmatians room because that was my partner's favourite movie as a kid. And I'm sure the only reason we got it was because it was a one-night stay as a lot of people were checking out (end of Thanksgiving weekend) so it was easy to slot us in.
