We booked an on-site trip with friends for the end of Sept. Their plans are changing and we are now likely going to stay off-site. I already booked our fastpasses for 60 days. Will I lose them now? Should I wait to cancel our on-site til the trip is with in 30 days or will I lose them no matter what?

I don't think you'd lose your FPs, regardless, but...
Edit-
(This assumes that you will have valid tickets to cover your entire trip in your MDX account that are not a part of your WDW resort package.)

You won’t lose your FPs if you cancel as soon as possible. By canceling now, you open up the onsite room for others to book.
