going for one day to Universal. Please provide thoughts on the following itenenary. Emphasis on best attractions and minimal lines...

Go to Studios first thing.
Get fastpass (or whatever its called) for Skrek
Go on Mummy

Go to IOA
do Hulk, Spidey, Dragon, then Jurassic

back to Studios
do Jaws, Earthquake, MIB, Back to Future

I assume you are not staying onsite. You've got the right plan.

The key is arriving early, the parking garage opens at 8am and a line of cars forms at 07:45am.

Your plan is perfect, just see Shrek when you arrive to USF. Don't worry about a fast pass for it.
Next ride the Mummy, if you really like it, and think you want to do it in the afternoon, get a fast pass now, otherwise, head over to IOA. The Mummy has a single rider line, so you may want to use that later.

Head left and ride Hulk. If you really like it, get a fast pass for noon. If you already got a fastpass from the Mummy, you won't be able to do this.
Spiderman (could user single rider line, but it can be long).
Dr. Doom (again, use single rider line)
Dueling Dragons
JPRA
Rip Saw Falls
Popeye

You may want to skip Rip Saw Falls and Popeye because you will get wet and it's not fun walking around a park, hot, sweaty, with wet cloths.

Head back to USF and do the shows. Use single rider line for MIB. Use your fastpass for the Mummy if you got one. BTTF will make you sick, it's one of those simulators that tricks your brain.

I disagree about doing Shrek first. That used to be my recommendation but not lately. If you enter the park at 8:45 when the gates often open, they wait to fill up the rooms before they start and you spend about 15 or so minutes wating. If you go at 9:00 AM, the lines in both express and regular line are very long. I waited for nearly 30 minutes in express line early in the morning. It was crazy. In fact the express line was as long as the regular line at that time. The shortest wait times for Shrek is right before park closing as almost everyone hits Shrek as one of their first rides.

I'd get an express for Shrek and pick a time that is the latest in the day so you don't have to rush back from IOA. Then I'd ride Mummy and head for IOA. MIB lines will be very long by the time you get back to Universal Studios so I suspect you may need to do single rider line for that.

My family of 4 is visiting Universal for the first time at the end of August. We are staying at Disney for 5-6 days first. Considering that we will have a guest asssistance pass for my autistic son & have minimal waits and that we will go on most non-rollercoaster rides and take pictures with characters , is it possible to get to both US and IOA in one day? I'd appreciate if anyone could help, especially a family with a similar situation.

The parks are pretty empty the end of August and with a guest assitance pass, and no crowds, and no rollercoasters, you should have no trouble seeing both parks.
