We are a group of 8 going to WDW and 1 of us will be in an ankle cast for the trip. I know that a GAC card is only good for 6 people total, so would they allow my friend in a wheelchair and her husband to go though the separate entrance and wait while the other 6 of us got through the line?

A GAC is for invisible disabilities. A wheelchair is visible-so no need for a GAC. Most lines at WDW are able to handle wheelchairs. Sue has a post in the sticky on how to best handle those that can't.
