The important thing is that your glasses fit well. Look down at the floor and shake your head like a wet dog drying off. If your glasses move around, get them adjusted.
I have a strap for my glasses, but I don't use it on anything at WDW. I did use it on RnRC the first time I rode it, but not anymore. If it makes you more comfortable to use one, you should, they're cheap and pretty comfortable.
Also, if your vision is so bad that you can't function without your glasses, you should travel with a spare pair (or at least a copy of your prescription). A roller coaster isn't the only bad thing that could happen to them. Ask me about the time I lost mine in the ocean or the time a drunk guy shoved me face-first into a door at a restaurant.
