Is Express Pass worth it for one day? Are lines usually very long on a Saturday in late August?
 
How's this for a good solid answer - it depends!

Everybody's definition of "very long" is different. As is the comment made by some - "seeing everything"

Are you trying to do both parks or just one? Are you a Potterhead? Are you just riding the main rides or trying to catch some of the shows as well? Do you have any interest in some of the smaller rides, like Seuss/Hippogriff/Woody etc.?

Late August usually starts to slow down, at least compared to early August, but it is a weekend. One thing for sure is Express Pass will not be sold out that day, you can get there, check things out, then decide to buy or not.

Universal does not take a lot of planning, but checking out a park map on the website might be a good start to at least have a mental checklist of what you need to accomplish to consider it a good day.
 
For late August, I'd try without. You can always buy it in the park if you decide you need it. The locals can increase park attendance on the weekends - but by August, I'd imagine they wouldn't want to spend all day in a hot park when it's going to eventually cool down.
 
They sell the pass at multiples locations inside the park, so you can go a judge if it's worth it or not, it will cost the same, I will recommend check the onsite Hotels that includes the pass and a early entrance to Harry Potter land on their nights rates it might work better. You will save also the parking fare when staying onsite
