I have a big Birthday trip coming up. 4 nights at the Poly and 4 nights at the Grand Floridian. During the poly stay I have a friend joining me for the first 2 nights, then they are leaving, and I have another friend joining for the last two nights. Will it be possible to have the resort remove that guest and add another guest for the last 2 days without incurring an extra charge? Thanks in advance.

I believe so, but it may end up being 2 separate bookings.
Call the main line with your booking info and see what they say. We have added 2 grandchildren to our room later in our stay without an issue, or having to pay more.
Good luck...

Yes, just speak with a CM at the front desk when your second guest arrives and explain the situation.

You do not need two separate reservations. Simply go to the front desk on the day your first guest is leaving and second guest is arriving and explain the situation. As long as you don't go over the two-adults-in-a-room rule, you will not be charged the extra adult fee and there is no reason to make two reservations.
