For us
Our top priorities are:
US
Minions
Gringotts
Exploring Diagon Alley
Eating at the Leaky Cauldron
IOA
The train over -- Universal to IOA -- the ride back is not on our list just the one over.
Forbidden Journey
The Jurassic Park River Ride and Discovery Center
**********************
Not must dos but given that the above doesn't take a full day we add in:
US -- The next tier for us on the unique offerings
Mummy
Men in Black
Transformers
Rip Ride Rocket
IOA -- nothing here you can't find something similar to at other parks, but we enjoy coasters and run of the mill coasters and amusement rides and do these if we have time
Hulk -- a decent coaster
Dueling Dragons
teacups if you can handle spinning -- for some reason unknown to me our college aged kids love these. They like the teacups at the MK too.
Dr. Doom -- drop ride -- better than Six Flags / lame compared to Cedar Point / lack theming like Disney's classic Tower or Terror
Spiderman is a must for some people and a unique IOA offering, but for us it's kind of a been there, done that, bought the t-shirt and we skip it.
