Can you remove days from a reservation once booked?

We are trying to plan a trip coming up and are undecided on total days. Can I book a 3 day trip now, and then call to remove one of the days if our plans change? Or do I have to cancel the entire reservation? This is assuming it is still within the modification period before a trip

There's no definitive answer to this question. If your resort/room category is available for the new dates, it is easy to modify. If it is not in the room inventory, you can try calling and asking them to modify your dates, but there is no guarantee they will do this.

Technically, you should be able to login to the site and modify your stay in the MDE section.

You should also be able to go the front desk during your stay and drop the nights you want to drop without penalty.

If you call, usually they will modify it for you. If the first CM says they can't, tell them to leave it as is. Call back and try again. Adding days is much more difficult than subtracting.

I just removed a night from our reservation through MDE last night. Very easy. I just went to our reservation, selected "change dates", changed to the new dates, and then at the top it gave me the option to stay in the standard room I had booked, or if I scrolled down, there was another room view I could choose.

Disney reservation system doesn't show the prices for individual days like many other hotels do. If you booked a certain number of days and got pricing associated with that, trying to remove days could change the room rate. While you can try to change it online or by calling, there is no guarantee you will be able to do that.

Sometimes a 4 night stay is available to book, but a 3 night stay (same dates) is not - presumably due to a booking algorithm meant to maximize occupancy. In this case, you can’t shorten the reservation by modifying online.

Yes, either via MDE or calling, you can easily remove a day from the beginning or end of your trip.
