I will be visiting Animal Kingdom for the first time in February with DH and DDs (almost 3 and 6 months). I have a few questions...
1. Is it possible to do the Safari ride and walk the Pangani trail before the 10am Lion King show if we head to the Safari at rope drop and have a fast pass for Lion King?
2. Will we be ok making our 10:45 Tuskar House reservation after the 10am Lion King show?
3. Which characters usually meet at the character outpost? Would it be likely we can get that for our 4th fastpass? My DD2 loves mickey and friends and we will probably try and see them everywhere we can on this trip.

Yes, definitely doable. Just keep an eye on the time as you are walking the trail.

Yes, you will be fine.

Mickey & Minnie and you might be able to get it as a 4th, but chances are you can catch it with a wait of less than 20 minutes if you are there on a recommended (not busy) day.
 
1. It's probably doable but I wouldn't. For one, even if you get on the safari right at 8:55-9am, it's still a 15 maybe 20 minute ride depending on how fast the driver is and if any animals hold you up. Then to walk the trail and really take it all in, you need about 30 minutes I'd say. We spend at least 15 of those minutes watching the gorillas. For FOTLK, we had a FP for the 10am show and lined up about 9:35, and we got good seats but those who walked in closer to 10am were in the back of the sections. There's not really a bad seat in the house but those on the ends of rows towards the front definitely got more interaction.

1 is theoretically possible. For perspective, last week my kids rode EE twice and Kali twice by 9:45. We had a safari FP at 10.

That doesn't mean it will work. You may find yourself rushing through the trail. I think the safari is manageable though.

As an FYI--you do take strollers in the line at safari and at a certain point, CMs will take them from you and park them in stroller parking. You won't know precisely where it is parked, so it may take a few minutes to find your stroller after Safari.

I don't have an answer for 2 or 3.

My kids are out of strollers now. But that is a good point. I remember at times trying to find ours after it was re-arranged by CMs.

I would try getting those two attractions in before your FP for the Lion King show. It's doable. But, it's your first time at AK. There are plenty of distractions. Animals on display (especially the talking birds), CMs holding insects in a box right after you enter, maybe DeVine, and so on. Plus, it's decent walk to the Safaris on uneven ground.

Be sure not to get distracted right after you walk in the entrance if you are pressed for time.

First thing in the morning for shortest wait. If you mean best times in terms of animal activity, it really varies. A lot of people say they are most active first thing in the monring, but we've not necessarily found that to be the case.
 
We did almost this exact schedule a few weeks ago. I have a DS8 and DS2 with stroller. We were in the bag check line right at 9 when the parked opened and had no real delays getting into the park. We headed straight to the safari (we weren't rushing but we didn't stop elsewhere) did that and then just made the 10am Lion King show standby. There is no way we would have had time to do the Pagani Trail. Then we had a 10:45 ADR at Tusker House which we made with no issues. It worked out really well, not a lot of rushing but again, we did not do the trail.
 
I've decided we are doing Safari right at opening, then racing to the 10am Lion King show, then racing to see if we can catch the 10:45Flights of Wonder (no big deal if we don't, we'll just hit the next show, but want to try!), and then we'll do the Nemo show, and I booked a 2nd Safari as a fp+ after lunch, my dd really wants to go on it twice. lol
