We are looking to get a Disneyland Deluxe AP with DVC discount. If our 1st use is on a block-out Saturday, can we get in that day?

Yes, if you activate your pass (first day of use) on a block out day, you can enter the park. That's what we did when we purchased ours last June. It was funny, the CM asked us if we would like to enter the park and we said, "um, YES!" We even parkhopped to DCA with no problem, we just mentioned that we activated the pass that day.
