They would cut off entry to all attractions for non-WDW-resort guests at the top of the closing hour (10PM, in this example.)
If (IF) Splash Mtn is operating for PM EMH:
For WDW resort guests, they would stop entry to the lines at (or a few minutes before) the end of the second EMH hour (12 Midnight, in this example.)

You can get in the line until the end of the posted operating hours. People not staying in the resorts can get in line for those rides at 9:59. People staying in Disney resorts can get in the line until midnight if that is the end of the EMH.
Bottom line: you can get in line until closing time and stay until you get your turn to ride. Even if it's a long line and it means you are riding well after the stated park closing time.

On certain rides, however, it is possible that if the CMs see that the line is way too long, they can close further entry to the queue at some point before the posted "closing time."
The "earlier" closing of some attractions and queues is more common during PM EMH.
