Two rooms help

We booked a second room today for family that is joining us. I linked their confirmation in my MDE and the travel agent noted our request to have connecting or adjoining rooms when she made the reservation. Is there anything else I can do to up our chances of having the rooms, at the very least, close together?

You didn't say which resort but if you are doing online check-in, then I suggest neither of you make any other requests, just leave it what the TA put in there. They will then know that is the most important thing.

If you are arriving together, you could skip online check-in and just do it at the front desk on arrival. If your rooms are not together, then you would have an opportunity to see if they can move you closer.

Nope, you did what you can. Just wanted to mention that you will still be fine even if you aren't close together. In January our oldest son ad his girlfriend discovered they were able to meet us there after we had made our reservations. There weren't any rooms left at our resort (we were just staying at ASMu in a Family Suite since our 16 year old son was with us as well) so they stayed at Sports. It wasn't a big deal, we just met them bright and early every morning and headed to the parks together.

I would not make any other room requests other than that. Adding other requests (upper floor, near transportation, specific buildings, close to pool, etc) lowers the possibility of getting the connecting rooms. Keep in mind that "adjoining" means that they will be near each other but not adjacent to each other.
