Yes. Well, you have to wait until the ship is cleared and it is announced that passengers leave the ship to get off and you have to get on before the all aboard times. But if you are asking if you can get off, do something in port, get back on the ship, and then leave the ship again to do something else in the port, then yes, you can do that.
 
To add to what Maggie'sMom said, if you are in a tender port, you will have to wait for tenders to be available. In Cabo San Lucas, we were told we may have to wait a while so that they could get all the Disney arranged tours off first, then we could go off on our own, but we were pretty much able to go right away, coming back, there was no wait. There was a RCL? ship also in port and their tender line was at least 100 yards long and 5 people wide. Disney had cold water and towels for us...they got nothing. It was sort of sad to see the comparison between the two and made me happy I was on the DCL ship.
