Quickest way to drive from Ft Meyers->Orlando?

We are visiting relatives in Ft Meyers, spending the night at a local Holiday Inn, then leaving late morning/afternoon to drive to Orlando. Mapquest says its about a 3.5 drive. What is the quickest way to get there and what is the best time of the day to drive? DH wants to say goodbye to his family in the AM before we go, so I don't know when we will be leaving. Thanks!
 
But I would assume going along Alligator Alley.

Head up to Tampa and then across.

I would think the easist way would be to go up interstate 75 towards Tampa and then get on interstate 4 to Orlando.
You could also take the more 'scenic' route and cut up through the middle of the state. Go up 75 until it hits route 17. Take route 17 north and it eventually hits interstate 4. It is shorter in miles, but is a local road at times with traffic lights and such. I did this once, in the opposite direction going from Orlando to Naples and it was an ok ride. :Pinkbounc

The quickest & easiest route is to take I-75 north. Then you pick up I-4 east and head to Orlando!
It's the way we always travel from Fort Myers to WDW!
Door to door it takes us a little over 3 hours.

We'll be driving this route next sunday morning! And we can't wait!!!

Having gone to Ft myers in 2000 to see a Sox game and visit friends! I Second going I-75-to I-4!!!
So much easier then the old days when you had to go over the sunshine bridge and use I-275
