Pay off balance with gift cards?

Hi all! I'm in Florida for a convention, and can't resist looking at rooms for this weekend. If I book a room only reservation for 2 nights (I know they will take the 1st night's pay now), can I pay the balance at check-in with gift cards? This would make the stay a lot more reasonable, as I already have enough gift cards to pay off at least one night's stay.
