If you decide to use this service, but stay at another hotel at the very beginning or end of your stay, can you still do it?

For example, if I make reservations as follows:

Night 1-Nickelodeon hotel
Nights 2-4 -Polynesian

Can I send our luggage to the Poly on that first day and they'll hold it for us (take a carry on to Nick Hotel.) Than, we can get it when we check in the next day.

Is that allowed?

I don't believe so - Magical Express is only for transportation to and from Disney resorts.

You could use Magical Express just for your return trip, and use a taxi or towncar or mears to take you to the Nickelodeon hotel.

No you would have to do the Poly stay first then the Nick hotel. You can't just use DME for luggage transfer and I would think there would be a problem with them holding your luggage if your check in date isn't until the next day.
