Is it in itself a sin?

No. It is not in and of itself a sin. For example. It is not a sin to be intolerant of murder. But intolerance in certain cases, such as that sign, can be a symptom and a result of sin, ie prejudice, bigotry, and outright hatred, all of which result in injustice.
