Going through the regular queue

This will be our first trip to Universal and we'll be staying onsite. I remember reading that you need to go through the entire queue on some of the rides like on Spiderman if you're doing them for the first time because it has something to do with the storyline. What are the other rides we need to do this for?
 
At Universal, the not to miss queues are:
Revenge of the Mummy (for the storyline) and Men in Black (just because it's cool). Mummy and MIB also have some interactive things in the line, if you know the secrets.

At Islands of Adventure:
Don't miss the queue in Spiderman (for the storyline), Dueling Dragons (awesome queue), Doctor Doom, and possibly Popeye and Dudley Do-Right (not for the storylines but because they are fun to see at least once).
 
spderman has a diffrent story for FOTL which is good as most fotl miss stories:jester:
 

We did the express line last year for everything that we went on, including Spiderman, and did not seem to miss anything. We all enjoyed all these rides (MIB, Spiderman, etc.)

Granted, the Mummy wasnt open yet, so i cant say for that one.

How would you know if you missed anything if you didn't do it? My first visit to Universal, we did FOTL for everything and so we missed all the queue. Our next visit, we didn't have FOTL and I was amazed at some of the queues, especially Dueling Dragons. I love FOTL but really some of the queues are worth waiting inline for.
