I am not sure you realize some of the logistics of this trip. Go to the homepage here and do some reading at the "theme parks" tab. If you are staying in Tampa you are at least an hour from orlando and maybe even closer to 2 hours each way from Walt Disney World. If your arrangements are already made in Tampa I would consider just doing Busch Gardens unless the girls are up to a long drive each day.

If you have changed your accomodations and are now staying in Orlando then I would suggest the Magic Kingdom if your only doing 1 day in the park. There really isn't anything in the Magic Kingdom that a 6yo can't do or wouldn't be tall enough except maybe space mountain (44" height restriction). As far as waiting in line its just part of the deal but you can use fastpass to avoid some of the lines. At 6 my girls loved all 4 parks so if you have time consider animal kingdom or MGM or a 2nd day at the magic kingdom.

Depending on your accomodations you could eat breakfast in the room and maybe pack some snacks and drinks but you won't want to carry a picnic around all day. If you stay on-site at disney you could return to the resort for lunch and maybe a swim.

In addition to this great site check out www.allearsnet.com. You will find more park touring information, tips for traveling with preschoolers and children as well as menus and ticket options.

Other than AAA and Alamo car rental and some on-line ticket vendors discounts on Disney tickets are very hard to find.

Well this one's easy! I don't know where you're staying in Tampa(it's a pretty big town) but get directions from the hotel to I-4 East. Then you just stay on I-4East until you hit Disney and follow the signs. Very, very easy to do! I imagine the drive will probably be just over 1 hour. Be sure and leave the hotel early enough to arrive before the parks open. Early morning is your least crowded time of day.

Has anyone used the strollers? Are they soft? Can they carry anything? Should we bring our own?
 

The one thing I want to suggest about the drive (being from the Tampa Area originally) is that you make sure and leave time for "Malfunction Junction" anyone from Tampa will know what i am talking about... Where I-275 meets I-4 is uasually a nightmare... I-275 is the main interstate that loops around Tampa. Give yourself about 30 extra minutes already built in to your time for it, if your directions take you on I-275 (depedning on where you are staying you might get lucky and not have to go through that area). If you hit any type of Rush hour that is where it will be at it's worst and there are always accidents there blocking up the road.

I would plan for an hour and a half to 2 hours drive time (there is a lot of construction going on on I-275 and I-4 that you may or may not have to contend with). But the drive itself is VERY easy and being that it is just really I-4 to Disney World it goes by pretty quickly. My family still lives in the Tampa Area and we drive it from WDW all the time.
 
I haven't rented the strollers, but I was talking in line with someone at the MK last month and she commented that the rental strollers don't have much storage. My lightweight stroller has a lot of storage and I liked having it since my 4yo was tired and wouldn't have wanted to walk to the car or bus leaving the parks.
