Personally unless it's to discuss a medical need for a specific location or something along those lines, I don't like the idea of calling the room controller. I think it's disruptive to their day. I prefer the idea of faxing. You can fax room requests, you don't need the agent to do so, although that's what they get paid to do.
Keep in mind that room requests are just that. Requests. Make sure to list things in the most important order. The more things you list the less likely you are to get what you want. It's best to list one or two, maybe three things.
If you get to your resort and really are disappointed with your room, you can always talk to guest services about moving rooms should something more to your desires open.
