Understanding that times can fluctuate a lot, I was wondering if I could get a rough sense of the average time to get from MCO to Gran Destino in the afternoon. Our flight arrives about 2:30 and I don't know if this historically tends to be a peak time or not.

Generally speaking, it's about 90 minutes from plane landing to arriving at your resort (if you're flying in domestically). Peak time doesn't really matter, when there are a lot of guests arriving they add more buses, and since DME is by reservation they do a pretty good job managing the logistics.
 
Disney recommends you allow 90 minutes for transportation. Please know that if it's not a "peak" time for Magical Express, the bus may (in my experience always) has more than one hotel to drop guests off at. You may be firs t resort, last resort or in between.

You also can be one of the first passengers on the bus and will sit and wait a few minutes for others to join you. (That's the reason when you book Magical Express they ask your flight number and arrival time).
 

Scenario one: You are last person on the bus, you are first hotel, it would be quick-ish.
Scenario two: You are first person on the bus, you wait about 10-15 minutes for bus to leave, CSR is last stop on the route, it would take longer.

Then there are tons of permutations in between.
 
I'd say anytime during the day is peak time. Late at night (like 10pm) is less crowded.
