Is it possible to get rooms together with connecting door in the Poly or the Caribbean?

Definitely. You need to make that request very clear on your reservation, explicitly stating "connecting rooms" (not "adjoining rooms" -- that just means next-door -- "adjoining" doesn't imply a door between the two rooms). Also, under most circumstances, recognize that it is just a request, and maybe you will get a connecting room or maybe you won't.

If you have small children with you, it's easier to obtain a connecting room.

Last year me and my DH stayed at CBR with my sister and 3 nieces in one room and a connecting door to our room, it worked great, except they are super early risers and would come running in to wake me and DH up at like 5 a.m.!!!!!!!! I am looking forward to our adult only trip this year!
