What is the best route to take from MCO to BWV? It'll be around 10am on a Saturday, so traffic shouldn't be too horrible.
 
When leaving the rental car garage follow the signs for the South Exit - Route 417. This will take you directly to Disney. Follow 417 to 536 (exit 3 I think). There will be two $1.50 tolls on 417. Rt. 536 will lead you right to Disney and once you go under the welcome to Disney sign just take the exit for the Epcot Resorts - think its the second exit. Stay in the right hand lane and when you get to the Hess station turn at the light and then turn left into the BWVs. Upon returning the car, there is a Hess station just before you get on 417 that is the closest to MCO for filling the car before returning.
 
The exit off of Rt 417 is exit 6, labeled International Dr.

Even though it's a little further, I recommend the next exit, (I think it's 3) that takes you through the MAIN gate, with the big entrance sign. It's a great first experience when driving into WDW. Plus you won't get stuck in all of the traffic that all meets at Exit 6, the International Drive Crowd, the Downtown Disney and theme park Crowd and the Disney resort crowd. Lots of traffic there, especially on the weekends.
 

I wasn't suggesting which exit to use, I was just clarifing the PP that thought the exit for Rt 536 was exit 3.
There also happens to be the same sign when entering Disney property from 536 (Epcot Center Dr), I wouldn't refer to it as a MAIN entrance, because there are similiar signs at several other locations.
There is also an additional $.50 exact change only toll at exit 3, so if the OP opts to use exit 3 make sure you have a couple of quarters.
 
North airport exit and left at first light.
Little over a mile is next light, two quick lefts onto 528 west.
(to pay toll, about a dollar, using cash, you need to use what looks like an exit ramp)
At end of 528 take I-4 west (southwest).
Take Epcot exit off I-4, heads northwest.
After going under welcoming arches, take second exit onto Buena Vista (west).
I think it is the 4'th light, Hollywood studios on the left, Boardwalk on the right. (Preceded by light with Victory on the left, nothing on the right.)

If you missed the Epcot exit (continuation of 536 west) and took the next exit (Osceola Pkwy) you would be going around three sides of a rectangle where Buena Vista was the fourth side.

The original entrance (which is still the largest) is still called Main Gate. If you missed both the Epcot exit and the Osceola exit you would reach 192 (Irlo Bronson) west and run right into Main Gate taking you onto World Drive which runs north-south. For most guests coming from the airport or downtown Orlando, the Epcot exit is shortest including going to Magic Kingdom.

If you did not come in that way you might want to go out Main Gate at a later date just to see its welcoming arches. South on World Drive, exit WDW, turn around at the first light and come back. World Drive intersects Buena Vista west of Boardwalk.
