I booked a co-worker and his family into a 2-bdrm lockoff at VWL for the week after Thanksgiving. The co-workers wife would rather have a dedicated but they are also concerned that her parents may not be able to make the trip.

Am I correct in saying that they have more flexibility with a lockoff unit in that if the parents have to cancel, then the co-worker could simply drop either the 1-bdrm or the studio and keep the other unit ? If they had a dedicated unit, then they would have no flexibility and would have to see if a 1-bdrm or studio was available if they wanted to dowsize.

There is no more flexibility with one than the other. If you book a 2BR lock off and want to drop the 1BR, then the way it works is MS will cancel your entire reservation. If there is someone on the waiting list for a studio, you'll have to get behind them (and anyone else) on the wait list. For that time of year, there could easily be a wait list especially since we're already within the 7 month window. So in that regard, there is no difference in getting a dedicated or non-dedicated.

Depending on how early in the process they cancel will determine how easy this will be to do. If it's a real possibility that they may drop out, it might be easier to book two studios or a 1BR and one studio. They might not be very close to each other, but that would provide a lot more flexibility if someone cancels. Of course, booking anything at VWL at this point for that time of year may well be a challenge.

You cannot simply drop the other portion, you have to cancel the 2 BR and rebook the 1 BR. A small point if the unit is available, a biggie if it's on a wait list for any days. What they could do is book a 2 BR AND a 1 BR and cancel what they don't need.
