We're going to WDW in early June and will have 2 people using ECVs in our group- my 89 year old grandmother and my aunt with MS. Can the Disney buses accomodate two ECVs at the same time? Do most of the buses have W/C lifts?

Yes.
Almost 100% of the buses are wheelchair/ECV accessible. Some have lifts and some have ramps. The buses with ramps are the newer buses and are most people consider them more easily accessible. Each bus can hold up to 2 wheelchairs or ECVs.
If there is more than one bus stop at your resort, sometimes one spot will already be occupied when the bus gets to your stop. If that happens, you can either split your party and have one group wait for the next bus. Or, you can wait for the next bus.
There is more information about buses in the disABILITIES FAQs thread near the top of the board.
