My family and my sisters families are planning to go to DC during easter weekend. My question is would they be open on easter sunday?

The museums are open every day except Christmas. This will be a very busy weekend for the museums.

"They" who? The Smithsonian museums should mostly be open (and busy) as noted above. Most of the monuments should be accessible although the little gift stores may not be. Congress etc will not be in session. I would not think you could get a White House tour either. Cherry blossoms *may* be starting but I think it will be too early so they won't be "open" either. National Cathedral will be PACKED - but beautiful.
