Does "check-in at port" refer to the building on land which you first go to before boarding the ship? Or does it refer to the check-in process once you're on board the ship?
To my understanding, there is a check-in process both on land and on board the ship, so I'm not sure where it's best to ask about a room upgrade.

There are a couple of things called "check in." One is online; you can (although you aren't required to, but I bet everyone does) check in online at the
DCL website weeks before your cruise. That's where you give some information about charging, etc, and print out a couple of forms to sign and bring to the port. But if you don't bring them, it doesn't matter. As long as you show up with your passport/birth certificate and payment method, they have all the forms you need at the port. But, checking in online makes life easier, and again I bet basically everyone does it. That is also where you can reserve some shore excursions if you want (you can also do them on the ship if you want to talk to someone about them and look at their handouts).
The second thing called "check in" is at the building at the port when you arrive. That's where you turn in the forms and get your "Key to the World Card." That's your room key/account charge card/ID card for getting on and off at the ports. You then hang around for a while until your turn to board, and you walk right on the ship. There is no more checking in to be done on the ship (well...you can visit the spa, kids clubs, shore excursion desk, and guest services for various reservations and event tickets, but not a part of checking in). Once you walk on the ship, all is well, and you are on vacation!
And, finally, the hope for a cheap room upgrade is done in the land checking building, after you complete all of your checkin process. That is the time when you visit the upgrade station (just ask any cast member where it is).
