Additional question, by requesting first available, won't I be subject to getting some of the worst located room? I'd rather wait for the perfect room than just take one to get in one. We spend a lot of hotel time on our trips.

Not necessarily. You'll either be getting a room that wasn't occupied the prior night, or a room where someone checked out at 5AM. Those rooms can be anywhere.
