What's the best strategy for rebooking onboard?

GO EARLY

The second they open. Know what you want (category and dates and open cabins) by doing your homework before you leave. Have an idea of the price by checking DCL's site and the competitors. Heck, have a quote from DU waiting in the wings; you can transfer back to them once on land.

IMO, if you do the above, you should be in/out of the line in 15 minutes tops. And feeling a lot lighter (cash-wise)

We had very good luck walking by when the desk was staffed on the second day of the cruise. The kids were in the clubs. Danielle was at the desk. There was no line.

AFAIK, everyone gets the same "deal" (although we didn't even attempt to negotiate), so other than being efficient, that's about it.

You will want to have some idea of the dates at category (and perhaps cabin) you'd like to book - although you get the discount if you move it, so its acceptable to "book anything"

So I guess they won't negotiate if you have a cheaper quote?
