We've stayed at both. POFQ is the smaller resort. It doesn't matter which building you stay in, they're all just a few mins from the busses/food court/pool etc.
POR is much larger. Its split into 2 sections, aligator bayou (which has a rustic theme) and magnolia bend (mansion style, think "gone with the wind")
At POR we stayed in Oak Manor, which was the first mansion style building after the pool.
POR has lots of landscaping and gardens which make it beautiful, though due to this it's much more spread out than the french quarter. Even though our building was close, it was about 5-10 mins walk to the front of the hotel.
If your going for 5 days i'd suggest POFQ. Its compact size will make it quicker for a short stay, without loosing the deep south charm.
You can always take a walk up the sassagoula river from POFQ to POR (around 20mins walk) if you fancy a look round. Its a nice relaxing walk on a night.
POFQ also had better entertainment in my oppinion. They had a jazz player in the bar which fitted in with the theme lovely. POR had a comedy piano player in the bar. It tended to get very noisy on a night there in the bar so you couldn't have a relaxing drink.
