On site express pass question

We are thinking about going Thanksgiving Weekend, and I'm sure it's going to be very crowded so we are thinking about staying on site to get the express pass. I want to make sure we are understanding this correctly. If we reserve a room for that Friday night, can we get there first thing Friday morning, drop of our luggage, and get our express passes? That way they are good all day Friday and all day Saturday for staying just 1 night on property?
