If you plan on only visiting resorts and casinos on the strip, probably not. It's still better to have one than to not if you want to go from say Wynn to Mandalay or Luxor to Mirage. They can be quite the walk, and with a car all of the parking decks are free.
I always get one because I often go off-strip, but always stay at a hotel on the strip. I find downtown gambling to be much more fun and fair, and I go there at least every other day. I also like to go to some of the luxurious off strip casinos like Red Rock and Green Valley Ranch, they're absolutely amazing and match anything on the strip except in size.
If you want to head to the Palms, Rio, or Hard Rock or anything like that, you'll most likely need a cab or a rental car.
