Yes its possible. Hotwire will from time to time show 2 bedrooms available. For example, a recent Hotwire win in Orlando showed a traveller getting 2 bedrooms at the Lake Buena Vista Resort Village & Spa. Be aware that Hotwire can be generous with their hotel ratings. This one was reported as 4 stars on Hotwire but it's only rated 3.5 star hotel on other sites.

Hotwire is experimenting with a new search results page that actually allows you to filter by amenity, and included in that is the option for 2 bedrooms. The only thing is, there's no guarantee you'll get their new page. I get it roughly have the time. The other half I get their old page which doesn't allow you to filter, so it just means a bit more looking.
